About Glygen Products

What is a TopTip?
A TopTip is a pipette tip with a fine slit at the bottom (width 1-2um) which permits liquids to pass through but retains the chromatographic media (20-30um) in the tip. This eliminates the need for a filter and, thus, dead volume. When operated, the tip is placed onto the end of a single or multi channel pipette. The sample is aspirated and then released through the tip to bind, wash and elute the analyte(s) of interest. An adaptor is available if you should you wish to centrifuge the sample. The TopTip is available in 3 volume ranges, 1-10uL, 10-200uL and 100-1000ul and a wide range of chromatographic media.

What is a NuTip?
A NuTip is a pipette tip for sample preparation in which the chromatographic media is directly embedded in the inner surface of the tip without polymers or glue, maximizing the surface area in contact with the sample. When operated, the tip is placed onto the end of a single or multi channel pipette. The sample is aspirated and then released through the tip to bind, wash and elute the analyte(s) of interest. The NuTip is available in 3 volume ranges, 1-10uL and 10-200uL and 10-200ul Large and a wide range of chromatographic media.

What is the difference between a NuTip and a TopTip and Spin Column?
The NuTip is a tip in which the chromatography media is directly attached to the inner surface of the tip without polymers or glue, maximizing the surface area in contact with the sample and eliminating the need for filters within the tip. The TopTip is a pipette tip with a fine slit at the bottom (width 1-2um) which permits liquids to pass through but retains the chromatographic material (20-30um) in the tip. This also eliminates the need for a filter and, thus, dead volume.

How do you spin a TopTip ?
The products are supplied with an adaptor should you wish to use a centrifuge.

Are TopTips available with robotic / automatic sample preparation systems?
Glygen offers tips for any automation system, as needed by the customer such as ABI Symbiot,
Sample Workstation for MALDI TOF Biospectrometry, Micromass 2700-MS and MassPREP, Sample Preparation Stations, Genomic Solutions ProMS, Sample Preparation Station, Packard BioSciences MultiPROBE and many more.

How are the NuTips & TopTips packaged?
The NuTips & TopTips are available in boxes of 96 of a single chemistry. It is possible to customize an order to include different types of tips in one package of 96 tips. Please call 1-800-742-5624 to discuss customization options.

Can I re-use the NuTips & TopTips?
The NuTips & TopTips are recommended for single use only and therefore should not be re-used.

Which brand(s) of pipettes do you recommend for use with the NuTips and TopTips?
NuTips work with most commercially available micro pipettes. TopTips come with syringes which can be used for pushing the sample through the tip.

How do I process larger (>200uL) samples through the TopTips?
The syringes supplied with the tips are sufficient for pushing several ml of sample through the TopTip. Simply fill the syringe with the sample and push it through the TopTip.

What are the specifications of the sorbents contained within the NuTips and TopTips?
See above responses (#1-8) regarding  NuTip, TopTip or Spin Column.

What protocols are available for the NuTips & TopTips?
Protocols are shipped with each order according to the size of the tip and its chemistry. They can also be provided upon request from Glygen at
sales@glygen.com.

What range of sorbents are available in the tips and what application areas are they used in?

Sorbent

Application Areas

C18

Hydrophobic peptides

C8

Hydrophobic peptides

C4

Hydrophobic peptides

Hypercarb

More hydrophilic peptides

Hypercarb+C18

Hydrophobic & Hydrophilic peptides

HILIC

Universal trapping of hydrophilic peptides, some proteins and other polar solutes; removal of salts, detergents, lipids & urea

Trypsin

Tryptic digestion of proteins

POROS WAX

Trapping & concentration of acidic proteins, removal of salts, detergents & urea

POROS SAX

Trapping & concentration of acidic proteins, removal of salts, detergents & urea

POROS SCX

Trapping & concentration of basic proteins, removal of salts, detergents & urea

Titanium Dioxide

Highly selective sample preparation – chelating agents such as hydroquionoline, phosphate peptides etc.

Which solvents should be used to condition the sorbents?

This depends upon the sorbent. Full procedures are outlined in the protocols for the product range.

What are the benefits of NuTips and TopTips over competing products?

The NuTip offers no resistance of flow, a problem reported in competing products. In addition, this product is offered in a wider range of chromatographic materials than any competing product. The binding capacity of the NuTip 1-10uL is similar and in some cases better, than competing products. There is no direct competition for the TopTip.

Can eluted product be stored for analysis at a later date?

Yes, elute the product into a clean vial and then seal. This should then be stored at an appropriate temperature until required

How can recoveries be improved when using the NuTips & TopTips?

Ensure that the solvents used for conditioning the sorbent and then washing and eluting are selected according to the phase of the tip. Sometimes, slow flow through the TopTip provides better recovery.

How do I use NuTips & TopTips?

This depends upon the tip size and the media contained within the tip. Please refer to the protocol for a particular product.

What is the chemical compatibility of tips?

NuTips & TopTips are compatible with the most commonly used solvents in protein purification. Please contact us at 1-800-742-5634 or sales@glygen.com with any queries regarding chemical compatibility.

What is the difference between NuTips and SPE spin columns?

NuTips & TopTips are designed for sample preparation on a very small scale (low microgram). The bed volume of the products is several orders of magnitude smaller than the bed weights contained within SPE spin columns.

What material is used to make the tips?

The tip itself is manufactured from polypropylene

When would I use the Hypercarb material over C18?

You would use the Hypercarb for extremely polar compounds that would not normally be retained on C18. A Hypercarb & C18 mix is offered for a compound that requires both types of retention.

How do I select the correct tip for my sample?

This depends upon the analytes of interest and the volume of your sample. Please refer to the application table below:

What is the lowest volume of sample that can be handled by the NuTip?

The volumes of NuTips are 1-10uL and 10-200uL. However, sample volumes as low as 100nL can be handled by the tips. For smaller volume, use Gel-loader NuTip or LC-Fiber.
